La pseudogenización del regulador transcripcional MarT, contribuye a la patogenicidad de Salmonella Typhi

Salmonella enterica serovar Typhimurium (STM) es considerado un patógeno generalista ya que infecta aves, ratones y humanos. Por otro lado, Salmonella enterica serovar Typhi (STY) es considerado un patógeno de hospedero restringido que sólo infecta al ser humano. A pesar de estas diferencias STM y STY comparten un 97% de identidad entre los genes que comparten. La transferencia horizontal y la pérdida de material genético han sido esenciales para la diferenciación de ambos serovares. La isla SPI-3 es un ejemplo del resultado de estos dos procesos. SPI-3 codifica – entre otros genes - para misL y marT. En STM, misL es requerido para la colonización instestinal de aves y ratones infectados oralmente. Además, se ha reportado que MarT regula positivamente a misL en STM, de manera directa mediante el reconocimiento de la secuencia consenso TNAAANNNNNTNAAA en su promotor. Por otra parte, en STY existen genes de SPI-3 que se encuentran como pseudogenes (marT es uno de ellos); mientras que otro marco de lectura, como es el caso de surV (que está involucrado en la resistencia a H2O2) está presente sólo en STY. Es interesante que la expresión heteróloga de marTSTM en STY regula negativamente a surV, a nivel transcripcional. Además, análisis transcriptómicos de STM al interior de macrófagos muestran que la expresión de marT incrementa 10 veces, reforzando la idea de que este gen está involucrado en la virulencia de STM. Por estas razones, el objetivo de este trabajo es encontrar y caracterizar nuevos genes regulados por MarT. Con este propósito, se realizó una búsqueda global in silico de la secuencia TNAAANNNNNTNAAA para identificar posibles blanco de MarT. Una vez seleccionados y validados esos genes, se confirmó que MarT está involucrado en una regulación negativa de STY1408, un gen con 98% de identidad a quimiorreceptores de la familia MCPs. Ensayos en placas de agar semisólido muestran que STY1408 está involucrado en motilidad tanto en STM como en STY. Infecciones de células HEp-2 con STM y STY mutantes en el gen STY1408 muestran un defecto en invasión, mientras que infecciones orales en ratón con mutantes nulas de STY1408 en STM son defectuosas para colonizar hígado y bazo. Con esto, se concluye que MarT participa en la regulación negativa de STY1408, un gen involucrado en la virulencia de Salmonella enterica.
Salmonella enterica serovar Typhimurium (STM) is considered a generalist pathogen since it infects birds, mouses and humans. On the other hand, Salmonella enterica serovar Typhi (STY) is considered a restricted host pathogen that only infects humans. Despite these differences, STM and STY share a 97% of identity between their shared genes. The horizontal transfer and the loss of genetic material have been essentials for differentiation of both serovars. The genomic island SPI-3 is an example that arose as a product of these two processes. SPI-3 encodes – between others genes – for misL and marT. In STM, misL is required for gut colonization of birds and mouse, orally infected. Futhermore, it has been reported that MarT positivetly regulates misL in STM, in a direct manner by recognizing the consensus sequence TNAAANNNNNTNAAA on its promoter. On the other hand, in STY there is genes of SPI-3 that are found as pseudogenes (marT is one of these); while other ORF, as is the case for surV (that is involved in the H2O2 resintance) is only present in STY. Interestingly, the heterolgous expression of marTSTM in STY negatively regulated surV, at the level of transcription. In addition, transcriptomics analysis of STM inside of macrophage shows that marT expression increase 10 folds, reinforcing the idea that this gen is involved in the virulence of STM. For these reasons, the objective of this work is to find and characterize new genes regulated by MarT. With this propose, we did a global in silico search of the TNAAANNNNNTNAAA sequence to identify possible MarT targets. Once selected and validated these genes, it was confirmed that MarT is involved in negatively regulates expression of STY1408, a gene with 98% of identity to chemoreceptors in the MCP family. Semisolid plate assays shown that STY1408 is involved in motility of STM and STY. Infection of HEp-2 cells with STM and STY mutant in STY1408 gene show a defect in invasion. Oral infection of mice with STY1408 null STM are defective to colonize liver and spleen. We conclude that MarT is involved in negatively regulates STY1408 gen involved in the virulence of Salmonella enterica.
