Este trabajo de investigación se enfocó en el estudio y revisión bibliográfica de las DNAzimas 8-17 y 10-23, las cuales son cadenas simples de DNA con funciones catalíticas. Estas enzimas aceleraran la velocidad de reacción de ruptura de una cadena de ARN.Las DNAzimas tienen múltiples aplicaciones tanto en terapias de silenciamiento génico, biosensores o en el campo de la nano biotecnología. Estas aplicaciones abren paso al interés científico de querer desarrollar nuevas tecnologías o promover mejoras en las ya existentes. Para esto, es crucial dilucidar el mecanismo de reacción empleado por las DNAzimas. La estructura secundaria de las DNAzimas 8-17 y 10-23 es bien conocida. Para la DNAzima 8-17 hay variados estudios funcionales y estructurales que han permitido ir construyendo el mecanismo que utilizaría esta enzima, aunque aún no se encuentra del todo resuelto. Sin embargo, el caso de la DNAzima 10-23, los estudios han estado enfocados en el desarrollo de aplicaciones y aún son muchos los puntos que se desconocen en torno sula relación función-estructura, lo que dificulta el mejoramiento racional de estas aplicaciones. En este trabajo se enfocó en el estudio bibliográfico para entender los conceptos de catálisis enzimática, en conocer y familiarizarse con las enzimas de DNA y las estrategias catalíticas que utilizan en sus mecanismos de reacción. Específicamente, enfocándose en los estudios realizados con las DNAzimas 8-17 y 10-23para establecer posibles comparaciones y encontrar similitudes en las estrategias catalíticas empleadas por ambas enzimas, tomando como referencia la más estudiada DNAzima 8-17
This research workis focused on the study and bibliographic review of the DNAzymes 8-17 and 10-23, which are single strandedDNAswith catalytic functions. These enzymes will accelerate the reactionrateof cleavageof an RNA chain. DNAzymes have multiple applications in gene silencing therapies, biosensors or in the field of nanobiotechnology. These applications open the way to the scientific interest of developingnew technologies or to promote improvements in the existing ones. To dothis, is crucial to understandthe reaction mechanism used by DNAzymes.The secondary structuresof the 8-17 and 10-23 DNAzymes arewellknown. For the 8-17 DNAzymes there are several functional and structural studies that have allowed to build the mechanism of reaction forthis enzyme, although it is not yet fully resolved. However, in the case of DNAzyme10-23, the studies have been focused mostly on the development of applications and there are many points that are unknown yet about its function-structure relationship, which makes it difficult to rationally improve itsapplications.In this work, Ifocused on acarry out a bibliographic study to understand the concepts of enzymatic catalysis, to know and get familiar with DNA enzymes and the catalytic strategies they use in their reaction mechanisms. Specifically, focusing on the studies carried out with DNAzymes 8-17 and 10-23 to establish possible comparisons and find similarities in the catalytic strategies employed by both enzymes, taking as reference the most studied DNAzyme 8-17.
