La periodontitis o enfermedad periodontal, es una de las enfermedades más
prevalentes en todo el mundo, es una infección inflamatoria que afecta a las estructuras de
soporte de los dientes y se caracteriza por la destrucción progresiva del ligamento periodontal
y el hueso alveolar que puede resultar en la pérdida de la pieza dental. Porphyromonas
gingivales es una bacteria anaeróbica Gram negativo considerado uno de los agentes
etiológicos más importantes implicados en el establecimiento y la progresión de la
En la microbiota oral las especies bacterias se encuentran manteniendo relaciones
sinérgicas y antagónicas, estas últimas incluyen sustancias que inhiben el crecimiento de
otros microorganismos, tales como productos metabólicos, sustancias tipo antibióticos,
enzimas bacterioliticas y bacteriocinas, estas son polipéptidos que poseen actividad
antimicrobiana y son producidas por un gran número de bacterias.
En esta tesis se purificó y caracterizó un elemento difusible tipo bacteriocina
producido por Staphylococcus epidermidis capaz de inhibir el crecimiento de
Porphyromonas gingivalis. .La purificación se realizó mediante HPLC-RF y MALDI-TOFMS obteniendo 2 peacks con actividad de masas 2732 Da y 5795 Da, este elemento difusible
fue resistente al tratamiento con temperatura y pH pero parcialmente resistente al tratamiento
con proteinasas. Nuestros resultados indican el aislamiento de un nuevo elemento difusible
tipo bacteriocina de origen Staphylococcal capaz de inhibir a Porphyromonas gingivales.
Periodontitis or periodontal disease is one of the most prevalent diseases in the world, is an infection that affects inflammatory supporting structures of the teeth and is characterized by progressive destruction of the periodontal ligament and alveolar bone which may result in the loss of the tooth. Porphyromonas gingivalis is a Gram-negative anaerobic bacteria considered one of the most important etiological agents involved in the development and progression of periodontal disease. In the oral microbiota species bacteria are maintaining synergistic and antagonistic relationships. The latter include substances that inhibit the growth of other microorganisms, such as metabolic products, such as antibiotic substances, bacteriolytic enzymes and bacteriocins, these are polypeptides having antimicrobial activity and are produced by large numbers of bacteria. In this work, purified and characterized diffusible element type produced by Staphylococcus epidermidis bacteriocin able to inhibit the growth of Porphyromonas gingivalis. Purification was performed by HPLC-RF and MALDI TOF-MS to afford 2 activity Peacks mass 5795 Da and 2732 Da, the diffusible element was resistant to treatment with pH and temperature but partially resistant to treatment with proteinases. Our results indicate the isolation of a new type diffusible element bacteriocin to inhibit staphylococcal origin to Porphyromonas gingivalis.
